Bupropion, an FDA-approved adjunctive medication for smoking cessation, is metabolized by cytochrome (CYP) P450 2B6 to hydroxybupropion, which is metabolized by CYP2B1 to inactive metabolites. These researchers investigated whether the CYP2B6*6 allele, which is associated with slower bupropion metabolism, has a greater bupropion response than CYP2B6*1, which is associated with normal bupropion metabolism.
The 326 subjects of this study were enrolled in a smoking-cessation treatment trial, were white, and had the alleles of interest. Subjects were treated with bupropion (dose increased to 300 mg/day at day 4) or placebo for 10 weeks and were told to quit smoking at week 2. Abstinence was assessed by self-report and breath CO levels.
