Because the pathogenesis of Alzheimer's disease (AD) includes an inflammatory response, many investigators have conducted observational studies to examine whether use of nonsteroidal anti-inflammatory drugs (NSAIDs) is associated with reduced risk for AD. These studies have yielded conflicting results, and many have had methodologic deficiencies.
In this prospective, population-based, cohort study from the Netherlands, researchers enrolled 6989 subjects (age, 55 or older) without dementia at baseline. During an average follow-up of 7 years, subjects underwent serial testing for dementia using standardized techniques; information about NSAID use was obtained from computerized pharmacy records.
