Pregabalin (Lyrica) is FDA-approved for treating patients with seizures, diabetic neuropathy, postherpetic neuralgia, and fibromyalgia. In a manufacturer-sponsored randomized trial, researchers assessed whether the drug affects cognition adversely.
Thirty-two healthy volunteers (age range, 18–55) received either pregabalin (titrated from 50 mg daily to 600 mg daily during 8 weeks, with an option to down-titrate if substantial side effects occurred) or placebo. The dose at 8 weeks was then continued for 4 additional weeks. Two pregabalin recipients (and no placebo recipients) dropped out because of side effects, and the remaining pregabalin recipients reached the maximal dose (300 mg twice daily).
