Cutaneous T-cell lymphoma (CTCL) exhibits a chronic, relapsing and remitting course with variable responses to several cytotoxic and biologic therapies. In an industry-sponsored, open-label, single-institution, phase II trial of vorinostat (Zolinza; suberoylanilide hydroxamic acid; SAHA), researchers in Texas enrolled 33 patients with refractory CTCL. At baseline, the median body-surface area with CTCL involvement was 45%, and the median pruritus score was 8 (on a 0- to 10-point scale). Mycosis fungoides was noted in 85% of patients, including 11 patients who had diffuse erythroderma or Sézary syndrome. Patients were heavily pretreated, and 29 had received one or more previous chemotherapies (median prior therapies, 5).
