Treatment options for patients with multiple myeloma have expanded during the past decade, with improved induction regimens and the availability of sequential therapies for those with relapsed or refractory disease. To compare two bortezomib (Velcade)-based combinations, Spanish investigators conducted a two-phase, multicenter, randomized, phase III trial involving previously untreated patients aged 65 and older.
In the first phase, 260 patients were randomized to 6 cycles of bortezomib (standard twice-weekly dosing in cycle 1 followed by once-weekly dosing in cycles 2–6) plus either thalidomide and prednisone (VTP) or melphalan and prednisone (VMP).
