Chronic lymphocytic leukemia (CLL) is a highly heterogeneous malignancy with a spectrum of prognostic subgroups identifiable by cytogenetic markers. In general, the presence of an isolated del(13q) correlates with slow disease progression and long survival, whereas CLL with del(11q) or del(17p) correlates with early progression and shorter survival. Although the molecular underpinnings of these karyotypic alterations are now identified — altered p53 function with del(17p) and dysregulated microRNAs (miR) with del(11q) and del(13q) — the interactions of the alterations, if any, and the roles that they play in pathogenesis have been unknown.
