Increasing the dose intensity of chemotherapy for newly diagnosed diffuse large B-cell lymphoma (DLBCL) has been proposed as a means to improve survival and cure rates.
To test this approach, investigators in the U.K. conducted a phase III, multicenter, prospective, open-label, randomized trial involving 1080 patients with DLBCL (93% with stage II–IV disease; 7% with bulky IA or IB disease). Patients received either standard-dose R-CHOP (rituximab plus cyclophosphamide, doxorubicin, vincristine, and prednisolone) administered every 21 days for eight cycles or R-CHOP administered every 14 days for six cycles followed by two cycles of rituximab.
