Locally advanced gastroesophageal adenocarcinoma has a high rate of distant recurrence, which has driven a shift in treatment toward perioperative systemic therapy instead of radiation-based regimens. In the industry-funded, phase 3 MATTERHORN trial, the addition of durvalumab, an anti–programmed death ligand 1 (PD-L1) antibody, to perioperative chemotherapy with fluorouracil, leucovorin, oxaliplatin, and docetaxel (FLOT) improved event-free survival and pathological complete response at 2 years’ follow-up. Now, the investigators report final results for overall survival.
