Although 5-fluorouracil and platinum-based chemotherapy is standard treatment for advanced gastric cancer, overall survival (OS) with this therapy is less than 1 year. Trastuzumab is the only targeted agent to improve survival when added to chemotherapy, but its use is limited to the minority of patients with human epidermal growth factor receptor 2 (HER2)-positive disease. Second-line chemotherapy with a taxane or irinotecan is superior to best supportive care (BSC), but it confers limited survival benefits.
