The Janus kinase 1 (JAK1) and JAK2 inhibitor ruxolitinib was shown in an early trial to reduce spleen size and improve symptoms in patients with intermediate- and high-risk myelofibrosis (MF; NEJM JW Oncol Hematol Sep 15 2010) and, in a follow-up to that study, to prolong survival in patients who continued full-dose therapy (NEJM JW Oncol Hematol Aug 14 2012).
