Ponesimod, which selectively and reversibly modulates the sphingosine 1-phosphate (S1P) receptor 1 (S1PR1), has been suggested as treatment for chronic plaque psoriasis. It reduces lymphocytes in circulation without affecting their survival or activation. Drug half-life is 30 hours; it is essentially eliminated from the body 1 week after the last dose. In a new, manufacturer-funded, phase II, placebo-controlled study, researchers (3 employed by the manufacturer) compared effectiveness and safety of ponesimod (20 mg or 40 mg daily) in 326 patients with moderate chronic plaque psoriasis.
