To compare two previously established standard-of-care myeloma regimens, investigators conducted an industry-supported, multinational, randomized, prospective, phase III trial involving 1623 patients with newly diagnosed myeloma not eligible for autologous stem cell transplantation. Patients were randomized to lenalidomide plus weekly dexamethasone (LD) for 18 months; continuous LD until disease progression; or melphalan, prednisone, and thalidomide (MPT) for 18 months. The primary endpoint was progression-free survival (PFS).
