The proteasome inhibitor bortezomib has single-agent activity in relapsed mantle cell lymphoma (MCL) and has been safely combined with rituximab immunotherapy and cytotoxic chemotherapy. Now, investigators have conducted an industry-sponsored, multicenter, randomized phase III trial to test whether substituting bortezomib for vincristine in R-CHOP therapy (rituximab, cyclophosphamide, doxorubicin, vincristine, prednisone) would be effective in previously untreated, non–transplant-eligible MCL patients.
