Waning immunity after vaccination potentially predisposes vaccinees to more-severe disease during infection with a new serotype, so long-term follow-up is essential. Investigators now present pooled efficacy data for year 3 of two phase III trials — and years 3 and 4 of one phase IIB trial — of CYD-TDV, a recombinant, live, attenuated tetravalent dengue vaccine (NEJM JW Infect Dis Dec 2014 and N Engl J Med 2015; 372:113). These industry-funded trials included >35,000 children aged 2 to 16 years in Asian-Pacific and Latin American countries; vaccine was administered in three doses (at 0, 6, and 12 months).
