Gastric cancer patients who experience progression after first-line chemotherapy achieve modest survival benefit from second-line treatment with taxanes and ramucirumab. Aside from HER2-targeted therapy, other agents targeting a biomarker-enriched population have not been identified. Investigators now report the results of an industry-sponsored, randomized, double-blind, placebo-controlled, phase II trial involving patients with advanced gastric cancer who received paclitaxel (80 mg/m2 on days 1, 8, and 15 every 28 days) plus either placebo or the poly (ADP-ribose) polymerase (PARP) inhibitor olaparib (100 mg twice daily).
