Although the appearance of atopic dermatitis (AD) is strongly influenced by environmental factors, predisposition to its development is determined by certain genes. For example, mutations of the FLG gene that encodes filaggrin result in deficiency of barrier function of the stratum corneum. Other susceptibility genes are being discovered via various strategies and models, including genome-wide association studies (GWASs) that compare genomes of affected and unaffected persons.
