In the landmark placebo-controlled GAIT trial of patients with knee osteoarthritis (NEJM JW Gen Med Mar 15 2006 and N Engl J Med 2006; 354:795), chondroitin sulfate and glucosamine hydrochloride, alone or in combination, were ineffective in the overall study population; however, an exploratory analysis suggested that combination therapy might benefit patients with moderate-to-severe knee pain. In this new double-blind, randomized trial from Europe, 606 patients with knee osteoarthritis and moderate-to-severe pain received combination chondroitin sulfate (400 mg) and glucosamine hydrochloride (500 mg) thrice daily or celecoxib (200 mg daily) for 6 months. The study was funded by a maker of chondroitin and glucosamine.
