For treating relapsed mantle-cell lymphoma (MCL), the Bruton tyrosine kinase inhibitor ibrutinib has been approved in the U.S. and Europe, and the mTOR (mammalian target of rapamycin) inhibitor temsirolimus is approved in Europe. Investigators compared the two drugs in a multinational, open-label trial funded by ibrutinib's manufacturer.
The researchers randomized 280 patients (median age, 68) with relapsed or refractory MCL to receive standard dosing of oral ibrutinib or intravenous temsirolimus. Patients had received a median of two prior lines of therapy, but not with either study drug. Participants with disease progression on temsirolimus were allowed to cross over to ibrutinib. Median follow-up was 20 months.
