Despite the success of agents that target vascular endothelial growth factor (VEGF) and epidermal growth factor receptor (EGFR) to improve survival in patients with metastatic colorectal cancer, once these therapies fail, survival is poor. Now, investigators have conducted an industry-supported, multicenter, open label, phase II trial to evaluate the potential benefit of the HER2-targeted combination of trastuzumab and lapatinib in patients with HER2-positive, metastatic, KRAS wild-type, chemotherapy-refractory colorectal cancer. The authors had previously established that this regimen had significant activity in HER2-positive, KRAS wild-type tumor xenografts.
