Investigators in Japan performed two industry-funded, phase III, noninferiority trials to compare the efficacy of the non–FDA-approved potassium-competitive acid blocker vonoprazan (20 mg) and lansoprazole (30 mg) for endoscopic healing of gastric ulcers and duodenal ulcers. They randomized 482 patients for the gastric ulcer study and 372 patients for the duodenal ulcer study. Endoscopy was performed at study entry and at 2, 4, and (for duodenal ulcers) 6 or (for gastric ulcers) 8 weeks.
In the duodenal ulcer trial, after 6 weeks, 94% of patients treated with vonoprazan and 94% treated with lansoprazole had complete ulcer healing. In the gastric ulcer trial, after 8 weeks, healing rates were 96% with vonoprazan and 98% with lansoprazole. Non…
