Emerging evidence indicates that right-sided colorectal cancers have different biology and are associated with a poorer prognosis compared with left-sided tumors. Investigators have now conducted a retrospective analysis of the prognostic and predictive impact of the primary tumor site in patients with metastatic RAS wild-type colorectal cancer who were treated in two recent industry-funded trials: the CRYSTAL study of therapy with FOLFIRI with or without cetuximab (NEJM JW Oncol Hematol Jul 2009 and N Engl J Med 2009; 360:1408) and the FIRE-3 study of therapy with bevacizumab versus cetuximab and FOLFIRI (NEJM JW Oncol Hematol Nov 2014 and Lancet Oncol 2014; 15:1065).
