Marginal zone non-Hodgkin lymphomas (MZL) are a heterogeneous group of B-cell malignancies characterized by extranodal involvement, association with specific antigenic dependency (e.g., Helicobacter pylori and gastric MZL), and a generally indolent clinical course. There is no standard therapy for relapsed or refractory patients.
To evaluate the efficacy and safety of the Bruton tyrosine kinase inhibitor ibrutinib in this setting, investigators conducted an industry-sponsored, multicenter, phase II trial of 60 relapsed or refractory MZL patients who had progressed after at least one anti-CD20–containing regimen (median number of prior systemic therapies, 2; range 1–9).
