Primary chronic cold agglutinin disease (CAD) is a clonal B-cell lymphoproliferative disorder accompanied by the production of monoclonal antibodies directed against the complement protein C3d, resulting in an autoimmune hemolytic anemia. Bendamustine is an alkylating agent approved for the treatment of indolent B-cell non-Hodgkin lymphoma and chronic lymphocytic leukemia. Rituximab is a monoclonal antibody against the B-cell protein CD20.
To examine the safety and efficacy of combination therapy with bendamustine and rituximab for CAD, investigators in Norway conducted a prospective, multicenter trial involving 45 CAD patients (median age, 74). Individuals with aggressive lymphoma and other malignant diseases were excluded.
