Certain comutations may have deleterious effects on survival and response to adjuvant chemotherapy in early-stage resected non–small-cell lung cancer.
Some of the most common oncogenes in non–small-cell lung cancer (NSCLC) are TP53, KRAS, and EGFR, but the prognostic and predictive effects of comutation status in early-stage NSCLC remain unknown. Investigators used the Lung Adjuvant Cisplatin Evaluation database in a meta-analysis of four adjuvant chemotherapy studies (3533 NSCLC patients) as well as mutation analyses of the resected specimens. In all, 1181 patients were included in the TP53/KRAS comutation analysis and 404 in the TP53/EGFR analysis.
In the TP53/KRAS analysis, 48% of patients had no known mutations, 32% had TP53 mutations, 16% had KRAS mutations, and 4% had TP53 and KRAS comutations. The TP53/KRAS comutation was not prognostic but was predictive of worse survival on adjuva…
