Investigators developed a messenger RNA (mRNA)-based vaccine encoding rabies virus glycoprotein (CV7201). They now report results of the first-in-human, open-label, prospective phase 1 clinical trial, supported by the vaccine manufacturer. Rabies was chosen because most humans are naive and correlates of immunity are known. The investigators enrolled 101 healthy, rabies-unvaccinated volunteers (aged 18–40 years), who received three doses intradermally or intramuscularly, via either needle-syringe injection or one of three needle-free devices, over 28 or 56 days. Subsequent cohorts received escalating doses. Fourteen participants received a booster dose at 1 year.
