No current treatment substantially improves negative symptoms in schizophrenia, and some medications produce effects that mimic negative symptoms. One promising molecule is MIN-101, a novel cyclic amide derivative with high affinities for sigma-2 and serotonin 2A (5-HT2A) receptors and binding affinity for alpha-1-adrenergic receptors, but low or no affinity for other receptors including pre- or postsynaptic dopamine (D2) receptors. In an industry-sponsored, phase 2b, international trial, researchers randomized 244 symptom-stable adults with schizophrenia to 12 weeks of MIN-101, at 32 mg or 64 mg daily, or placebo.
