Ruxolitinib is a JAK1/2 inhibitor approved for the management of myeloproliferative disorders, including myelofibrosis and hydroxyurea-resistant polycythemia vera. To evaluate its safety and effectiveness versus best available therapy (BAT) for essential thrombocythemia (ET) unresponsive to other therapies, Harrison and colleagues conducted a randomized, phase II trial involving 110 ET patients resistant or intolerant to hydroxyurea.
In the ruxolitinib group, the median dose received in the first year was 19 mg twice daily. In the BAT group, 71% of patients received hydroxyurea, 48% received anagrelide, and 40% received interferon.
