Associations between proton-pump inhibitors (PPIs) and a host of medical problems, including stroke, have been suggested in retrospective database studies. Because of the nature of these studies, causation cannot be proven. Now, investigators have evaluated the possible association between PPIs and stroke using data from two large prospective cohort studies. Over 97,000 participants (70% women) were followed for 12 years.
After adjustment for demographics, risk behaviors, and some medical factors, there was a slight association between PPIs and ischemic stroke (hazard ratio, 1.18; 95% confidence interval, 1.02–1.37), but not overall stroke or hemorrhagic stroke.
