Dupilumab (Dupixent) is an anti–interleukin (IL)-4 receptor monoclonal antibody that inhibits IL-4 and IL-13 signaling and reduces type 2 inflammation. In two industry-funded studies, investigators examined the efficacy of add-on therapy with twice-monthly dupilumab injections in patients (age, ≥12) with asthma.
In one trial, 210 patients with oral corticosteroid–dependent asthma were randomized to receive dupilumab or placebo for 24 weeks. Mean reduction in steroid dose, while maintaining asthma control, was 70% in the dupilumab group and 42% in the placebo group; dupilumab resulted in significantly fewer estimated annual exacerbations (0.6 vs. 1.6) and a forced expiratory volume in 1 second (FEV1) that was higher by a mean 0.22 L.
