In a prior phase II study, the second-generation, irreversible, epidermal growth factor–receptor tyrosine kinase inhibitor (TKI) dacomitinib was shown to significantly prolong progression-free survival when compared with the first-generation TKI erlotinib in patients with advanced non–small-cell lung cancer (NSCLC; NEJM JW Oncol Hematol Nov 2012 and J Clin Oncol 2012 Sep 20; 30:3337).
Now, investigators have conducted an international, randomized, open-label, phase III study (ARCHER 1050) to compare overall survival (OS) with dacomitinib versus the first-generation TKI gefitinib in 452 patients with treatment-naive metastatic EGFR-mutated NSCLC.
