Studies in mice have revealed that high blood levels of glucose stimulate production of thioredoxin-interacting protein, which promotes death (apoptosis) of insulin-producing β-cells, and that verapamil attenuates production of thioredoxin-interacting protein.
Twenty-four people with recent-onset type 1 diabetes were randomized to once-daily verapamil (≤360 mg) or placebo, along with insulin therapy, for 1 year. β-cell function was assessed by measuring levels of insulin C-peptide following a standardized meal. During 1 year, the verapamil group exhibited superior β-cell function, smaller mean increase in total daily insulin requirement (27% vs. 70%), and fewer hypoglycemic events (0.5 vs. 2.7 events/month), compared with the placebo group. …
