Overall survival trended longer with the addition of palbociclib to endocrine therapy in patients with ER-positive/HER2-negative metastatic disease.
Randomized clinical trials combining a CDK4/6 inhibitor and endocrine therapy in patients with ER-positive/HER2-negative metastatic breast cancer have uniformly demonstrated impressive and clinically meaningful improvements in progression-free survival (PFS) with both first and later lines of therapy. However, improvements in overall survival (OS) have yet to be reported.
One recent industry-sponsored, international, randomized, double-blind, placebo-controlled, phase III trial reporting PFS (PALOMA-3; NEJM JW Oncol Hematol Aug 2015, and N Engl J Med 2015; 373:209) involved 521 patients with ER-positive/HER2-negative metastatic breast cancer who progressed or relapsed during prior endocrine therapy. At a median follow-up of 5.6 months, PFS w…
