Despite the availability of new targeted therapeutic agents for acute myeloid leukemia (AML), durable disease control remains poor among older patients.
To evaluate the use of maintenance azacitidine in this setting, investigators conducted an industry-supported, multicenter, randomized, phase III trial (HOVON97) involving 116 older AML patients (median age, 69 years; range, 60–81) who achieved complete remission (CR) or CR with incomplete blood count recovery (CRi) after two or more cycles of intensive induction chemotherapy. Patients were randomized to observation or azacitidine (50 mg/m2 on days 1–5 every 28 days until relapse or a maximum of 12 cycles). About half of patients were able to complete 9 to 12 cycles of azacitidine.
