The M72/AS01E candidate vaccine against Mycobacterium tuberculosis is composed of a recombinant fusion protein, M72, derived from two M. tuberculosis antigens along with the AS01E adjuvant. Previously, a placebo-controlled phase IIb trial was undertaken in three African countries on the efficacy of two intramuscular doses of the vaccine in preventing pulmonary tuberculosis in HIV-negative adults (aged 18–50) with latent tuberculosis based on a positive interferon-gamma release assay. A 2-year interim analysis showed 54% protective efficacy (NEJM JW Infect Dis Nov 2018 and N Engl J Med 2018; 379:162).
