Hypertrophic cardiomyopathy (HCM) can cause symptoms of heart failure, either from abnormal diastolic left ventricular (LV) function or from LV outflow obstruction in patients with the obstructive phenotype (oHCM). In a phase 2 study (NEJM JW Cardiol Jul 2019 and Ann Int Med 2019; 170:741), the non–FDA-approved reversible inhibitor of cardiac-specific myosin, mavacamten, substantially reduced LV outflow gradients in oHCM. Now, researchers report on a manufacturer-sponsored, placebo-controlled, dose-ranging, phase 2 trial assessing adverse events with mavacamten for symptomatic nonobstructive HCM (nHCM; MAVERICK-HCM; NCT03442764).
