Researchers analyzed combined data from three large, ongoing prospective health studies to determine any association of development of type 2 diabetes with use of proton-pump inhibitors.
Among over 200,000 patients free from diabetes who were followed for a median of 8 to 10 years, 10,105 developed type 2 diabetes. Diabetes risk was increased in PPI users (hazard ratio, 1.24). The majority of excess risk occurred among patients who used PPIs for >2 years, and risk increased with greater duration of use. Although PPI users were more likely than nonusers to have several chronic conditions, the analysis took that into account and confirmed the independent association.
