The authors conducting this study sought to develop a fast, reliable assay for α-synuclein-containing extracellular vesicles in Parkinson disease. There were 170 patients with Parkinson disease and 131 healthy controls.
Lumbar punctures were performed to obtain cerebrospinal fluid analysis. The α-synuclein was quantified using antibodies against both the total and aggregated fractions. A nanoscale flow cytometry technology was applied. The total α-synuclein and aggregated α-synuclein-positive extracellular vesicles were lower in patients with Parkinson disease than in controls. The combination of the two extracellular vesicle subpopulations yielded a sensitivity of 80% and a specificity of 71% for the test.
