With the advent of next-generation sequencing in colorectal cancer, targetable mutations have been identified, including DNA mismatch repair protein deficiency, BRAF V600E mutation, NTRK gene fusion, and HER2 overexpression. Recent studies have indicated activity for HER2-directed agents in HER2-amplified or -overexpressing colorectal cancer.
Investigators report the results of DESTINY-CRC01, an industry-sponsored, open-label, multicenter phase 2 trial of trastuzumab deruxtecan, an HER2 antibody–drug conjugate, in HER2-expressing metastatic colorectal cancer.
