The addition of an antimyeloma therapeutic monoclonal antibody to regimens containing an immunomodulatory agent, a proteasome inhibitor, or both, plus weekly dexamethasone, improves responses in patients with multiple myeloma. Investigators report an international, prospective, industry-sponsored phase 3 trial comparing carfilzomib plus weekly dexamethasone with or without the addition of the anti-CD38 monoclonal antibody isatuximab (10 mg/kg weekly for 4 weeks followed by every 2 weeks). Adult patients who had received one to three prior lines of therapy were randomized 3:2 to the isatuximab arm (n=179) or the control arm (n=123).
