Bispecific therapeutic antibodies are active in relapsed B-cell malignancies by dual engagement of a tumor cell surface antigen and CD3 on T-cells, thus juxtaposing tumor and effector cells. In a phase 1, multicenter, industry-funded trial, investigators evaluated intravenous or subcutaneous administration of single-agent teclistamab, a bispecific antibody targeting the B-cell maturation antigen (BCMA) and CD3, in patients with relapsed or refractory multiple myeloma. The study's aims were to determine safety and tolerability of the agent and to identify the recommended phase 2 dose (RP2D). Treatment was continued until disease progression or unacceptable toxicity.
