In the AMBITION-cm study, a phase 3 randomized trial in Uganda involving persons with HIV and cryptococcal meningitis (NEJM JW Infect Dis Mar 24 2022 and N Engl J Med 2022; 386:1109), mortality at 10 weeks was similar after a single 10-mg/kg dose of liposomal amphotericin B (L-AmB) followed by 2 weeks of oral flucytosine and fluconazole as compared with a 7-day course of deoxycholate AmB at 1-mg/kg daily with flucytosine followed by fluconazole for an additional 7 days. The single-dose L-AmB regimen was endorsed by the World Health Organization (WHO) in 2022.
