In a 2022 observational study from Denmark, researchers reported that offspring of men who took metformin during the 3 months before conception (the period of sperm development) were statistically more likely to have congenital malformations than were children whose fathers were unexposed to metformin (NEJM JW Gen Med May 15 2022 and Ann Intern Med 2022; 175:665). However, a subsequent cohort study from Israel did not confirm the Danish findings (NEJM JW Gen Med Jul 15 2024 and Ann Intern Med 2024; 177:851).
