Vaccines aimed at the developmental stage of Plasmodium falciparum can affect the parasite's growth in mosquitoes, potentially decreasing community transmission. In further testing of the Pfs230D1 vaccine against P. falciparum gametes, investigators randomized 236 participants in Mali (mean age, 30 years) to 4 injections of high-dose (Pfs230D1 full), fractional dose (Pfs230D1 fractional), or comparator vaccine. In addition to testing the vaccine's safety and immunogenicity, efficacy was assessed by measuring the number of infected mosquitoes in a direct skin feeding (DSF) assay 6 weeks after the third dose.
Both the full and fractional doses of Pfs230D1 were well tolerated, with the most common reactions being injection site pain and fever. …
