Several medications are now FDA-approved to treat obesity — with more in the pipeline — but limited head-to-head data exist to guide treatment decisions. To address this, researchers conducted a network meta-analysis of 260 randomized trials of obesity medications involving nearly 100,000 participants (median age, 49; median body-mass index, 35). Follow-up was a median of 26 weeks.
Compared to lifestyle interventions, tirzepatide produced the most weight loss (≈15% of body weight).
Oral semaglutide, injectable semaglutide, orforglipron, and phentermine-topiramate each conferred weight loss of ≈10% of body weight.
