Use of proton-pump inhibitors (PPIs) during pregnancy is thought to be safe, but good data are lacking. In this meta-analysis, Canadian researchers examined seven cohort studies that included 1530 women who were exposed to PPIs (during at least their first trimesters) and 133,000 unexposed controls.
PPI use was not associated with elevated risk for major congenital malformations, spontaneous abortions, or preterm delivery. The same findings held for omeprazole specifically (which accounted for 87% of the PPI exposures in this study).
Comment
Although analyses like this one cannot necessarily capture very rare adverse outcomes, the findings suggest that PPIs can be used safely during pregnancy.
